In site-directed spin labeling (SDSL), a stable radical, usually a nitroxide, is covalently attached at a specific location within a macro-molecule, and structural and dynamic information at the labeling site is obtained via electron paramagnetic resonance (EPR) spectroscopy. My group pioneered the development of a family of nitroxide labels, called the R5-series, which can be attached in an efficient and cost-effective manner to a specific phosphorothioate backbone position at arbitrary DNA or RNA sequences. Using the R5-series, we have obtained unique information on nucleic acids and protein-nucleic acid complexes in bulk solutions and at the single-molecule level. I will present an overview of our work with a focus on studying sequence-dependent DNA deformation by the p53 tumor suppressor and investigating DNA unwinding by CRISPR-Cas.
